چکیده
Detailed Methods Cell Culture The human THP-1 macrophage-like cell line was obtained from American Type Culture Collection (ATCC, Manassas, VA). For experiments, cells were cultured at 4.0x10 cells/35-mm plate (Falcon Scientific, BD Biosciences) in RPMI 1640 supplemented with 10% fetal bovine serum (Sigma), β-mercaptoethanol (5x10 mol/L), 100 units/mL penicillin, and 100 μg/ml streptomycin and differentiated with 300 nmol/L phorbol 12,13-dibutyrate (PDB, Sigma) for 1 week prior to use in experiments as described. HepG2 cells were obtained from ATCC and grown as described previously. For experiments, HepG2 cells were plated in either 100-mm or six-well (35-mm) culture plates (Falcon, Mississauga, ON) and maintained in minimal essential medium (MEM) containing 10% fetal bovine serum (FBS), but switched to MEM containing 5% human lipoprotein-deficient serum (LPDS) for experimental incubations.

In vitro and in vivo assessment of inhibitory effect of stevioside on pro-inflammatory cytokines
Objective: Stevioside is a natural non-caloric sweetener which has been reported to have anti-inflammatory activity. The aim of the present study was to examine in vitro and in vivo effects of stevioside on rats plasma levels of tumor necrosis factor- α (TNF-α), interleukin-1β (IL-1β), TNF-α and IL-1β release from lipopolysaccharide(LPS)-stimulated rat peripheral blood mononuclear cells (P...
متن کاملEffect of Surgical Flap on IL-1β and TGF-β Concentrations in the Gingival Crevicular Fluid of Patients with Moderate to Severe Chronic Periodontitis
Background: Growth factors play a major part in wound healing in many tissues including the periodontium. Transforming growth factor-β1 (TGF-β1) is one of these factors present in the gingival crevicular fluid. In addition, it is considered as one of the most important anti-inflammatory cytokines. Interleukin-1β is a proinflammatory cytokine that presents itself in gingival inflammation and the...
متن کاملCorrelation between NF-κB signal pathway-mediated caspase-4 activation and Kawasaki disease
The aim of the study was to investigate the role and mechanisms of action of nuclear factor-κB (NF-κB)-mediated caspase-4 activation in the induction of inflammatory cytokines during Kawasaki disease (KD) and coronary artery endothelial cell injury. Peripheral blood mononuclear cells (PBMCs) were isolated from KD patients and healthy controls and cultured. Double antibody sandwich enzyme-linked...
متن کاملSingle nucleotide polymorphisms associated with P2X7R function regulate the onset of gouty arthritis
BACKGROUND Gout is an inflammatory disease that is caused by the increased production of Interleukin-1β (IL-1β) stimulated by monosodium urate (MSU) crystals. However, some hyperuricemia patients, even gouty patients with tophi in the joints, never experience gout attack, which indicates that pathogenic pathways other than MSU participate in the secretion of IL-1β in the pathogenesis of acute g...

Objective(s):In the present study, we evaluated immunological and immunomodulatory properties of royal jelly (RJ) in 2,4,6 trinitrobenzene sulfonic acid (TNBS)-induced colitis in rats. Materials and Methods: Eighteen adult female Wistar albino rats were divided into three groups of six animals each: a control group that received only saline solution, a TNBS-induced colitis group, and a TNBS-co...
